As well as playing them in Agents of S.H.I.E.L.D., Iain De Castecker and Elizabeth Henstridge reprised their roles as Fitz and Simmons respectively in the animated Ultimate Spider-Man (4.5) vs The Sinister Six: Lizards.
However I do not think that this established a continuity between the live-action Agents of S.H.I.E.L.D. and the animated Ultimate Spider-Man.
In fact Simmons in the Ultimate Spider-Man is a blonde and that is hardly her hair colour in Agents of S.H.I.E.L.D.
The Fitz and Simmons actors are not the first from Agents of S.H.I.E.L.D. to play their characters in Ultimate Spider as Clark Gregg voiced Phil Coulson during the first two seasons of the animated series which started not long before he reprised Coulson for Agents of S.H.I.E.L.D.
